Angiogenesis Assays

Stem cells have demonstrated potent angiogenic activity that can be both beneficial and detrimental. Angiogenesis is often a desirable and clinically relevant outcome in cell therapy paradigms where cardiovascular ischemic damage results or where vascularisation is required. On the other hand, cancer stem cells can progress tumor formation through stimulation of blood vessel formation. Whether your research is focused on induction or inhibition of stem cell mediated angiogenesis, AMSBIO offers optimized kits, supporting reagents and cell lines required to address your research needs.

In Vivo angiogenesis DIVAATM assay*

To recapitulate the true in vivo environment, angiogenesis sprouting from existing vasculature, AMSBIO offers the only in vivo angiogenesis kit available. The Directed In Vivo Angiogenesis Assay (DIVAATM) kit permits in vivo angiogenesis analysis within a controlled silicon cylinder, called an angioreactor. The sleek design of the patented angioreactor optimizes sensitivity and allows for up to four units per mouse, giving a more controlled experiment and decreasing the number of mice required for a study. DIVAATM is available as a Starter Kit, Inhibition Kit and Activation Kit. Angioreactors can also be filled with 3D matrix of your choice, including MAPTrix Hygel TM.

In Vitro Angiogenesis Assays

Angiogenesis is a multi-step process involving the sprouting and growth of new blood vessels from existing vasculature. In multiple in vivo models, stem cells have been shown to modulate angiogenesis in response to injury, disease and cancer. The initial stages of angiogenesis involve recruitment (invasion & migration) of endothelial cells followed by tubule formation and maturation. In vitro angiogenesis assays provide a controlled environment to focus on individual stages of the angiogenic process. In vitro assays are designed to study endothelial cell invasion and tubule formation. Stem cells and/or compounds modulating angiogenesis are easily introduced into the assay resulting in a quantitative assessment of pro- or inhibitory angiogenic bioactivity.

In vitro angiogenesis tube formation assay

Cultrex® In Vitro Angiogenesis Assay Kit provides a costeffective method for investigation of prospective stem cell mediated angiogenesis in a 96 well plate format. The Tube Formation Assay Kit contains reduced growth factor Basement Membrane Extract qualified for low levels of tube formation in the absence of angiogenic factors and high levels of tube formation in the presence of angiogenesis factors, using human umbilical vascular endothelial cells (HUVEC). Endothelial cells are available from AMSBIO (not supplied in the assay kit), see Angiogenesis Tools and Reagents (p41) for details. The assay kit includes a tested inhibitor as a negative control and supplies both colorimetric and fluorometric cell staining reagents

During the in vitro Tube Formation Assay endothelial cells (1) directionally migrate to align (2), branch (3) and form polygonal networks (4).


Endothelial cell invasion assay

During angiogenesis, endothelial cells invade through the basement membrane to form sprouting vessels, which in turn invade injured or hypoxic tissue. This kit offers a flexible, standardized, 96 well high-throughput format for quantitating endothelial cell invasion, employing a simplified Boyden chamber-like design with an 8 micron polyethylene terephthalate (PET) membrane. A control is provided for inhibition of in vitro endothelial cell invasion and detection is quantified using Calcein-AM.

In vitro vascular permeability assays

Vascular permeability can be a limiting critical property regarding toxic effects of cytokines and molecular exchange between vascular and tissue compartments. The Cultrex ® 96 and 24 well Vascular Permeability Assays accelerate the screening process for pathways and compounds that influence vascular permeability. These assays offer a flexible, standardized, high-throughput format for the quantification of the degree to which stem cells, genes or compounds influence the maintenance of endothelial cell-to-cell adhesion.

Endothelial progenitor cells

Human endothelial progenitor cells (EPCs) are isolated from samples using a proprietary technology and provided in cryopreserved or live flasks. Human endothelial cells are available from human aorta, umbilical vein, coronary and pulmonary arteries and dermal microvascular tissue. Donors have been screened and detailed donor profiles are available upon request.

Human Endothelial Progenitor Cells: Endothelial Progenitor Cells (Cat Nr Z7030071) in culture (passage 6)are double-positive for LDL uptake and and Lectin binding. Fixed cells were stained for: Dil-Ac-LDL (red), FITC-Ulex-Lectin (green) and DAPI nuclear counterstain (blue)

Media and supporting culture reagents

Endothelial progenitor and endothelial cell growth media are designed and tested to promote attachment, spreading and proliferation of human endothelium progenitor and endothelial cells. These media are available fully supplemented with fetal bovine serum and growth factors or serum-free defined medium is also available, with or without growth factors, to fully support angiogenesis assays. Each lot of medium is performance tested.

Angiogenesis antibodies

Angiogenesis antibody markers are available, including von Willibrand factor and CD31 antibodies to detect mature vessels and endothelial tubes, and many more.


Angiogenesis specific antibody microarrays

Each Angiogenesis Antibody Array includes 127 highly specific and well-characterized antibodies in the angiogenesis pathway; each has six replicates. The arrays are printed on standard-size coated glass microscope slides and can be scanned on all microarray scanners that are compatible with 76 x 25 x 1 mm (3 in. x 1 in. x 1mm)

The angiogenesis antibody microarray: Supplied as two identical subarrays (one subarray for normal sample and another for the treated sample). Each subarray contains 6 replicates for each antibody. Positive controls and negative controls are included in the array.

Transwell Cell Invasion and Migration Assays

The ability of stem cells to "regenerate" injured tissue is a powerful application of stem cells. Conversely, cell migration and invasion can also lead to disease progression that includes cancer, atherosclerosis and arthritis. Cell invasion is intimately related to cell migration, except invading cells move through the extracellular matrix (ECM), through degradation and proteolysis of the ECM, into neighboring tissues. AMSBIO offers cell invasion and migration assays in a variety of formats and extracellular matrices to suite your research needs. Cell Invasion and Migration assays are developed based on two technologies, the Boyden chamber and zone exclusion. CultureCoat ® Assay Kits employ the Boyden chamber and are available ready-to-use pre-coated with extracellular matrix; whereas ORISTM cell invasion and migration assays employ zone exclusion technology.

CultreCoat® ECM pre-coated cell invasion kits

CultreCoat® 96 Well ECM Coated Cell Invasion Kits offer a ready-to-use and standardized approach for providing optimal and accelerated results for screening stem cell bioactivity, compounds and pathways that influence cell migration through extracellular matrices. Employing a modified Boyden chamber-like design with an 8 micron polyethylene terephthalate (PET) membrane, the CultureCoat® Cell Invasion Assay Kits are available in 24 well and 96 well formats precoated with Basement Membrane Extract (BME), Laminin I, Collagen I or IV. The CultreCoat ® BME Cell Invasion Assay has been adapted to three different matrix coating densities, low, medium and high densities, as all cell lines do not exhibit the same invasive capacity. Sampler Kit offers low, medium and high BME densities for assay optimization. The coating process assures uniform layers of extracellular matrix plate to plate and lot-to-lot, to minimize variability.


These assays offer a flexible, standardized, high throughput format for quantitating the degree to which invasive cells penetrate a barrier consisting of basement membrane components in vitro in response to stem cell chemoattractants and/or inhibiting compounds. CultreCoatTM ECM Pre-Coated Cell Invasion Assay Kits are provided as a complete kit with rehydration solution, cell wash buffers and calcein AM.

Different coating densities for the 96 Well BME Cell Invasion Chamber result in different invasion rates for invasive cell lines. HT-1080, human fibrosarcoma and MDA-MB-231, breast cancer, cell lines were serum starved for 16 hours and seeded at 25,000 cells per well in pre-coated 8 m chambers. Cells invaded in response to 10% FBS over a 24 hour period and were quantitated using Calcein-AM. Samples were run in quadruplicate.

Zone Exclusion Cell Invasion and Migration Assays

ORISTM assays create a defined cell-free Detection Zone to quantifiably measure cell migration or invasion efficiently and easily.

ORISTM cell migration assays

One assay commonly used to study cell migration is the scratch assay; creating a cell-free gap, or "scratch", on a confluent cell monolayer. Whilst this method is popular, the inherent variability in how scratches are produced and physical damage to cells make it difficult to obtain quantitative and statistically valid measurements of cell migration. Furthermore it has been demonstrated that the scratch will damage any underlying extracellular matrix. Utilising cell stoppers or a biocompatible gel to create the cell-free zone, the unique design of ORISTM assays generates highly reproducible results using a microscope, digital imaging system or fluorescence plate reader. With no Boyden chamber insert needed, cell migration can be monitored in real-time without the need to fix cells and provides formats to cater for high throughput applications.

Cell Proliferation Assays

Optimized and reliable in vitro proliferation of stem cells is fundamental to obtain the desired quantity and quality of stem cells for your research. Common methods for determining cell viability depend upon membrane integrity (e.g. trypan blue exclusion) or incorporation of nucleotides during cell proliferation (e.g. BrdU or 3H-thymidine). These methods are limited by the impracticality of processing large numbers of samples or by the requirement for handling hazardous materials. The Calcein-AM Cell Viability Kit, XTT and MTT Cell Proliferation Assays provide rapid, versatile and accurate methods for quantifying cell viability in standard culture conditions. 3D Culture Cell Proliferation Assays are available that provide a more physiologically relevant assessment of stem cell proliferation within a 3D extracellular matrix.

Calcein-AM cell viability kit

The Calcein-AM Kit provides a simple, rapid and accurate method to measure cell viability and/or cytotoxicity. Calcein-AM is a non-fluorescent, hydrophobic compound that easily permeates intact, live cells. The hydrolysis of Calcein-AM by intracellular esterases produces calcein, a hydrophilic, strongly fluorescent compound that is wellretained in the cell cytoplasm. Cells grown in black-walled plates can be stained and quantified in less than two hours.

XTT cell proliferation assay

The use of XTT tetrazolium salt to assay cell proliferation, cell viability and/or cytotoxicity is a widespread, established practice. The procedures avoid radioactivity, allow for rapid determination in microplates and give reproducible and sensitive results. XTT, a yellow tetrazolium salt, is cleaved to a soluble orange formazan dye via the succinatetetrazolium reductase system in the mitochondria of metabolically active cells, which can be measured by absorbance in a microplate reader. This kit includes both XTT and the electron coupling reagent for a conven-


ient and simple assay.

MTT cell proliferation assay

MTT Cell Proliferation Assay is a sensitive kit for the measurement of cell proliferation based upon the reduction of the MTT tetrazolium salt. In actively proliferating cells, an increase in MTT conversion is spectrophotometrically quantified. Conversely, in cells that are undergoing apoptosis, MTT reduction decreases, reflecting the loss of cell viability.

3D culture cell proliferation assays

3D culture conditions provide the most accurate representation of the in vivo environment. The Cultrex® 3D Culture Cell Proliferation Assays offer a flexible, standardized, high-throughput format for quantitating cell proliferation within in vivo-like environments. The Cultrex® 3D Culture Cell Proliferation Assay has been adapted to multiple formats so that cell proliferation may be evaluated within different extracellular matrices (BME, Laminin I and Collagen I); and is available in a 96 well plate format.

Cell Adhesion Assays

AMSBIO offers Cell Adhesion Assays to evaluate optimal stem cell attachment to extracellular matrices and assessing factors that influence cell-matrix interactions. CultreCoat® Cell Adhesion Assays are pre-coated and ready -to-use in a black stripwell plate format that minimizes background, allows greater sensitivity and affords flexibility in the number of samples assessed. The stripwell feature also allows multiple experiments to be conducted simultaneously using the same kit. Calcein labeling allows direct comparisons between the number of cells that are loaded and the number that adhere. Controls are provided for determining background and non-specific binding.

Adhesion protein array kit

With this kit, on one plate, you can assess adhesion on Laminin, Collagen I, Collagen IV, Fibronectin and Vitronectin. Ideal for research applications that involve screening multiple different extracellular matrices.

Supplied as one 96 well stripwell plate pre-coated with the extracellular matrix of your choice and all the reagents required to conduct the assay.

Cell Adhesion Assay. Cells are pre-labeled with Calcein-AM, and loaded onto stripwell plates precoated with ECM. The cells attach to the ECM proteins, and total cells loaded is determined by fluorescence output. The unattached cells are then washed away, and the cells that have attached is determined by fluorescence output. Cell attachment is then calculated by dividing the fluorescence value for cells loaded by that of cells attached.

Human embryonic stem cell lines expressing surfactant protein ­ C (SP-C+)

Human ES cell lines are available expressing SP-C that present a primary human lung cell phenotype. In addition to SP-C protein, these cells express Alpha Fetal Protein (AFP), an early endoderm marker; Cytokeratin, a late endoderm marker; and NKX2.1, a endoderm and lung related marker. These SP-C+cells are suitable for Drug Metabolism Studies and Pulmonary Disease Models. Available from BG01Vderived and HUES-7V-derived human embryonic stem cell line backgrounds. Human embryonic stem cell derived astrocytes

Astrocytes are differentiated from the BG01V human Embryonic Stem Cell line using a proprietary method. The differentiated astrocytes are characterized based on their morphology, expression of Glial Fibrillary Acidic Protein (GFAP) and functional ability to support primary neurons. These cells are suitable for in vitro drug screening assays, toxicological predictive modeling for drug discovery and neurological disease models.
